1.in the tree
In the tree 表示的在树上指的是原本不在树上的物体，例如：鸟，猫等小动物，物品及人。
There are some birds in the tree.
树上有一些鸟。
The kitty is in the tree.
那个小猫在树上。
2.on the tree
On the tree表示原本在树上的东西，比如树枝、树叶、花、果实等。另外，其他一些明显位于树木表面，但不属于数本身的东西，也用on the tree。
There are some pears on the tree.
树上结了一些梨。
Those flowers in the tree come out.
树上的花开了。
